Standardisation of EGFR FISH in colorectal cancer: results of an international interlaboratory reproducibility ring study

چکیده
Aims Epidermal growth factor receptor (EGFR) gene copy number evaluated by fluorescence in situ hybridisation (FISH) can discriminate among KRAS wild-type patients those with better outcome to EGFR-targeted therapy in metastatic colorectal cancer, further enhancing selection of patients. Nevertheless, enumeration of gene copies is challenging and the lack of analytical standardisation has limited incorporation of the test into the clinical practice. We therefore assessed EGFR FISH interlaboratory consensus among five molecular diagnostic reference

Standardization of EGFR FISH in colorectal cancer: Results of an international, interlaboratory reproducibility ring study.
3597 Background: Epidermal growth factor receptor (EGFR) gene copy number (GCN) evaluated by fluorescence in situ hybridization (FISH) has been shown to discriminate among KRAS wild type patients those with better outcome to EGFR-targeted therapy in metastatic colorectal cancer (CRC), further enhancing selection of patients. Nevertheless, enumeration of gene copies in CRC is challenging and the...
متن کاملEvaluation of Expression of EGFR, HER-2 and COX-2 in Colorectal Cancer
Background and objective: EGFR and HER-2 are two members of ERbB/HER family of Type I Transmembrane growth factor receptors. Cox2 is an enzyme responsible for the conversion of arachidonic acid to prostaglandins, which has a major role in angiogenesis and can modelate tumor growth. The aim of this study was to determine the level of expression of EGFR, HER-2 and Cox2 in colorectd cancer. Mater...
متن کامل“How should we treat older patients with Metastatic Colorectal Cancer, A Review”
Nearly 50 % of newly diagnosed colorectal cancer, affect people over 70 years of age. Inclusion of older patients in clinical trials has been extremely rare. As a result, there is debate on how to manage these patients because it is still unclear how to balance the therapeutic advantages and toxicities. For patients who do not have comorbid conditions, with performance status (P.S.) 0–1, treatm...
متن کاملارزیابی فراوانی جهشهای ژن KRAS در بیماران ایرانی مبتلا به سرطان کولورکتال
Background: Kirsten rat sarcoma (KRAS) gene is a target of genetic alterations which are diagnostic and prognostic biomarkers in patients with metastatic colorectal cancer who are treated with monoclonal anti-EGFR antibodies such as cetuximab and panitumumab. KRAS mutations are seen in 35-42% of patients with colorectal cancer. The high frequency of these mutations in colorectal cancer represen...
متن کاملKRAS and BRAF mutations in Iranian colorectal cancer patients: A systematic review and meta-analysis
Background: Mutations in the EGFR signaling pathway play an important role in the development of colorectal cancer (CRC). Mutations in these genes, like KRAS and BRAF, affect the treatment strategies and associated with poor prognosis and relative resistance to anti-EGFR therapies. Our aim was to conduct a systematic and meta-analysis on all studies that have been conducted on the prevalence of...
